The first Donnybrook was the Hume Highway village now called Kalkallo, settled in 1840 with an inn by 1841, first known as Rocky Water Holes and renamed in 1853, probably after the Dublin parish. When the railway opened in 1872 the station and a new village 3 km east of the highway took the Donnybrook name, and the old one became Kalkallo in 1874. A post office came in 1889. The histories also list a cheese factory, a mineral spring with picnic and cabin facilities, and Donny Spa Park from 1997. The old cemetery is in Kalkallo, beside a patch of indigenous grassland. A 1,120 ha farm near Kalkallo was bought in 2011 for a mixed-use estate with a new station.

The English Street plan was approved in 2016 and the Donnybrook-Woodstock and Northern Quarries plans in 2017, the middle one by far the largest. Olivine, Peppercorn Hill and Katalia are three of the estates on them, with Heartford and Acclaim the others. Donnybrook Primary opened in 2023, Hume Anglican Grammar has a campus, and the murnong Community Centre opened in February 2026. Lawns here are mostly young turf on the basalt plain, cut higher and less often while it knits in. V/Line trains to Seymour and Shepparton stop at the station, and buses 501, 511 and 525 run through.

City of Whittlesea says residents and owners are legally required to mow the nature strip beside their land and keep the grass, weeds and scrub on it down. Every residential property received a 240 L bin, light green lid, for food and garden waste in June 2026, charged on the rates from 1 July 2026 rather than by an opt-in fee. It goes out fortnightly, in the week recycling does not, the same day the weekly rubbish goes; glass is collected every four weeks.
